    Background: In this work we predict enzyme function at the level of chemical mechanism, providing a finer granularity of annotation than traditional Enzyme Commission (EC) classes. Hence we can predict not only whether a putative enzyme in a newly sequenced organism has the potential to perform a certain reaction, but how the reaction is performed, using which cofactors and with susceptibility to which drugs or inhibitors, details with important consequences for drug and enzyme design. Work that predicts enzyme catalytic activity based on 3D protein structure features limits the prediction of mechanism to proteins already having either a solved structure or a close relative suitable for homology modelling. Results: In this study, we evaluate whether sequence identity, InterPro or Catalytic Site Atlas sequence signatures provide enough information for bulk prediction of enzyme mechanism. By splitting MACiE (Mechanism, Annotation and Classification in Enzymes database) mechanism labels to a finer granularity, which includes the role of the protein chain in the overall enzyme complex, the method can predict at 96% accuracy (and 96% micro-averaged precision, 99.9% macro-averaged recall) the MACiE mechanism definitions of 248 proteins available in the MACiE, EzCatDb (Database of Enzyme Catalytic Mechanisms) and SFLD (Structure Function Linkage Database) databases using an off-theshelf K-Nearest Neighbours multi-label algorithm. Conclusion: We find that InterPro signatures are critical for accurate prediction of enzyme mechanism. We also find that incorporating Catalytic Site Atlas attributes does not seem to provide additional accuracy.     Over the years, traditional active infrared thermography has played a pivotal role in ensuring that a component is free of any damage. However, whilst optical thermography is still not sufficiently sensitive to the presence of material micro-flaws, current vibro-thermography requires inspection solutions that are not always feasible, such as the use of coupling materials between the sensing probe and the monitored structure. This paper presents a valid alternative to current thermographic systems by developing and experimentally validating nonlinear aircoupled thermosonics for a contactless, rapid and accurate detection of fatigue micro-cracks in an aero-engine turbine blade. The proposed thermographic method combines the high sensitivity to micro-damage of nonlinear ultrasonic techniques with non-contact air-coupled ultrasonic transducers and thermographic equipment. Narrowband frequency sweeps were performed to identify local damage resonance frequencies in order to generate large vibrational amplitudes at the damage location and compensate for signal losses caused by the high acoustic impedance mismatch between the air and the sample. An infrared camera was then used to acquire the thermal response generated by frictional heat at the crack interfaces. Moreover, an image processing method based on a combination of morphological opening and a Savitzky-Golay smoothing filter was employed to enhance the quality of thermal images affected by anisotropic heating and thermal noise effects. Nonlinear aircoupled thermosonics experiments were validated with laser Doppler vibrometry scan measurements and compared with both flash and pulsed phase thermography. Thermal imaging results showed that the proposed nonlinear air-coupled thermosonics was the only thermographic technique able to detect fatigue micro-cracks, thus demonstrating its potential as an efficient and sensitive inspection tool for micro-damage detection in geometrically complex components.     The relationship between postprandial peptides at circulating physiological levels and short-term appetite control is not well understood.The purpose of this study was first to compare the postprandial profiles of ghrelin, glucagon-like peptide 1 (GLP-1), and peptide YY (PYY) after isoenergetic meals differing in fat and carbohydrate content and second to examine the relationships between ghrelin, GLP-1, and PYY with hunger, fullness, and energy intake.Plasma was collected before and periodically after the meals for 180 minutes, after which time ad libitum food was provided. Simultaneous ratings of hunger and fullness were tracked for 180 minutes through phases identified as early (0-60 minutes) and late (60-180 minutes) satiety.This study was conducted at the Psychobiology and Energy Balance Research Unit, University of Leeds.The participants were 16 healthy overweight/obese adults.Changes in hunger and fullness and metabolic markers were indicators of the impact of the meals on satiety.Ghrelin was influenced similarly by the 2 meals [F(1, 12) = 0.658, P = .433] and was significantly associated with changes in hunger (P.05), which in turn correlated with food intake (P.05). GLP-1 and PYY increased more by the high-fat meal [F(1, 15) = 5.099 and F(1, 14) = 5.226, P.05]. GLP-1 was negatively associated with hunger in the late satiety phase and with energy intake (P.05), but the PYY profile was not associated with hunger or fullness, nor was PYY associated with food intake.The results demonstrate that under these conditions, these peptides respond differently to ingested nutrients. Ghrelin and GLP-1, but not PYY, were associated with short-term control of appetite over the measurement period.

    The Whittard Canyon is a branching submarine canyon on the Celtic continental margin, which may act as a conduit for sediment and organic matter (OM) transport from the European continental slope to the abyssal sea floor. In situ stable-isotope labelling experiments were conducted in the eastern and western branches of the Whittard Canyon, testing short-term (3–7 days) responses of sediment communities to deposition of nitrogen-rich marine (<i>Thalassiosira weissflogii</i>) and nitrogen-poor terrigenous (<i>Triticum aestivum</i>) phytodetritus. <sup>13</sup>C and <sup>15</sup>N labels were traced into faunal biomass and bulk sediments, and the <sup>13</sup>C label traced into bacterial polar lipid fatty acids (PLFAs). Isotopic labels penetrated to 5 cm sediment depth, with no differences between stations or experimental treatments (substrate or time). Macrofaunal assemblage structure differed between the eastern and western canyon branches. Following deposition of marine phytodetritus, no changes in macrofaunal feeding activity were observed between the eastern and western branches, with little change between 3 and 7 days. Macrofaunal C and N uptake was substantially lower following deposition of terrigenous phytodetritus with feeding activity governed by a strong N demand. Bacterial C uptake was greatest in the western branch of the Whittard Canyon, but feeding activity decreased between 3 and 7 days. Bacterial processing of marine and terrigenous OM were similar to the macrofauna in surficial (0–1 cm) sediments. However, in deeper sediments bacteria utilised greater proportions of terrigenous OM. Bacterial biomass decreased following phytodetritus deposition and was negatively correlated to macrofaunal feeding activity. Consequently, this study suggests that macrofaunal–bacterial interactions influence benthic C cycling in the Whittard Canyon, resulting in differential fates for marine and terrigenous OM.

    Support Force Glacier (SFG) is a large ice stream feeding into the Filchner Ice Shelf. The active seismic survey is recorded at the sheet-shelf transition of SFG. We map the bed while grounded, the ocean cavity around a surface channel at the ice shelf and its counterpart at the base, the basal channel, and the seabed. The survey consists of 5 seismic reflection profiles, 2 along-flow profiles and 3 across-flow profiles. The 2.42GB seismic data were recorded with a 300 m long streamer consisting of 96 30Hz p-wave sensors. The sample rate is 0.5 ms, record length 3000 ms.     The impacts of ocean acidification on coastal biofilms are poorly understood. Carbon dioxide vent areas provide an opportunity to make predictions about the impacts of ocean acidification. We compared biofilms that colonised glass slides in areas exposed to ambient and elevated levels of pCO2 along a coastal pH gradient, with biofilms grown at ambient and reduced light levels. Biofilm production was highest under ambient light levels, but under both light regimes biofilm production was enhanced in seawater with high pCO2. Uronic acids are a component of biofilms and increased significantly with high pCO2. Bacteria and Eukarya denaturing gradient gel electrophoresis profile analysis showed clear differences in the structures of ambient and reduced light biofilm communities, and biofilms grown at high pCO2 compared with ambient conditions. This study characterises biofilm response to natural seabed CO2 seeps and provides a baseline understanding of how coastal ecosystems may respond to increased pCO2 levels.     AbstractTo have epidemic potential, a pathogen must be able to spread in human populations, but of human-infective RNA viruses only a minority can do so. We investigated the evolution of human transmissibility through parallel analyses of 1755 virus genome sequences from 39 RNA virus genera. We identified 57 lineages containing human-transmissible species and estimated that at least 74% of these lineages have evolved directly from non-human viruses in other mammals or birds, a public health threat recently designated “Disease X”. Human-transmissible viruses rarely evolve from virus lineages that can infect but not transmit between humans. This result cautions against focussing surveillance and mitigation efforts narrowly on currently known human-infective virus lineages and supports calls for a better understanding of RNA virus diversity in non-human hosts.

    The genus Paracallisoma (Crustacea: Amphipoda) is revised and the type species, Paracallisoma alberti is redescribed based on holotype material supplemented with new material from the region of the type locality. This revision results in the establishment of two new genera, Pseudocallisoma gen. nov. and Haptocallisoma gen. nov., and the description of a new species of Haptocallisoma and a new species of Paracallisoma from the North Atlantic Ocean. An account of all known species within the three genera is given and updated keys to the genera and species are provided.

    Background: In this work we predict enzyme function at the level of chemical mechanism, providing a finer granularity of annotation than traditional Enzyme Commission (EC) classes. Hence we can predict not only whether a putative enzyme in a newly sequenced organism has the potential to perform a certain reaction, but how the reaction is performed, using which cofactors and with susceptibility to which drugs or inhibitors, details with important consequences for drug and enzyme design. Work that predicts enzyme catalytic activity based on 3D protein structure features limits the prediction of mechanism to proteins already having either a solved structure or a close relative suitable for homology modelling. Results: In this study, we evaluate whether sequence identity, InterPro or Catalytic Site Atlas sequence signatures provide enough information for bulk prediction of enzyme mechanism. By splitting MACiE (Mechanism, Annotation and Classification in Enzymes database) mechanism labels to a finer granularity, which includes the role of the protein chain in the overall enzyme complex, the method can predict at 96% accuracy (and 96% micro-averaged precision, 99.9% macro-averaged recall) the MACiE mechanism definitions of 248 proteins available in the MACiE, EzCatDb (Database of Enzyme Catalytic Mechanisms) and SFLD (Structure Function Linkage Database) databases using an off-theshelf K-Nearest Neighbours multi-label algorithm. Conclusion: We find that InterPro signatures are critical for accurate prediction of enzyme mechanism. We also find that incorporating Catalytic Site Atlas attributes does not seem to provide additional accuracy.     Over the years, traditional active infrared thermography has played a pivotal role in ensuring that a component is free of any damage. However, whilst optical thermography is still not sufficiently sensitive to the presence of material micro-flaws, current vibro-thermography requires inspection solutions that are not always feasible, such as the use of coupling materials between the sensing probe and the monitored structure. This paper presents a valid alternative to current thermographic systems by developing and experimentally validating nonlinear aircoupled thermosonics for a contactless, rapid and accurate detection of fatigue micro-cracks in an aero-engine turbine blade. The proposed thermographic method combines the high sensitivity to micro-damage of nonlinear ultrasonic techniques with non-contact air-coupled ultrasonic transducers and thermographic equipment. Narrowband frequency sweeps were performed to identify local damage resonance frequencies in order to generate large vibrational amplitudes at the damage location and compensate for signal losses caused by the high acoustic impedance mismatch between the air and the sample. An infrared camera was then used to acquire the thermal response generated by frictional heat at the crack interfaces. Moreover, an image processing method based on a combination of morphological opening and a Savitzky-Golay smoothing filter was employed to enhance the quality of thermal images affected by anisotropic heating and thermal noise effects. Nonlinear aircoupled thermosonics experiments were validated with laser Doppler vibrometry scan measurements and compared with both flash and pulsed phase thermography. Thermal imaging results showed that the proposed nonlinear air-coupled thermosonics was the only thermographic technique able to detect fatigue micro-cracks, thus demonstrating its potential as an efficient and sensitive inspection tool for micro-damage detection in geometrically complex components.
