    ABSTRACT This work presents the preliminary result of the multidisciplinary cruise EXPLOSEA2 surveying the northern Mid-Atlantic Ridge and Azores Archipelago from 46◦ 30′ N to 38◦ 30′ N aboard the R/V Sarmiento de Gamboa and ROV Luso over 54 days (June 11 to July 27, 2019). In this cruise report, we detail the geophysical, hydrographic, geological, oceanographic, ecological, and microbiological data acquired and a brief of main findings. The cruise addressed the exploration and comprehensive characterization of venting sites, including the water column, the sediments and rocks that host the hydrothermal activity, and the associated mineralizations, biology, and microbiology. Deep hydrothermal chimneys and massive sulfide deposits (up 3,000 m in depth) within the Moytirra hydrothermal active field were identified on slopes that had not been explored previously. Another striking finding made during the EXPLOSEA2 cruise was the field of carbonate chimneys named the “Magallanes-Elcano” field, a potentially relict ultramafic-hosted hydrothermal site sourced by abiotic methane. This field is related to a serpentinite and gabbro rock outcropping on a dome-shaped massif named the “Iberian Massif.” An outstanding finding of the EXPLOSEA2 survey was the identification of the first garden of soft corals growing after active submarine eruptions were reported in the Azores Archipelago composed by a high density of soft corals the suborder Alcyoniina at the summit and flanks of a recent volcanic cone at 160 m water depth developed during the 1957–1958 eruption of Capelinhos. Several cold-water coral habitats formed by colonial scleractinians (e.g., Lophelia pertusa and Madrepora oculata), coral gardens composed of mixed assemblages of black corals (Leiopathes sp.), and octocorals and dense aggregations of the glass sponge Pheronema carpenteri that may be classified as vulnerable marine ecosystems (VMEs) have been discovered during the EXPLOSEA2 cruise along the northern Mid-Atlantic Ridge. This work reveals the importance of multidisciplinary surveys to the knowledge of deep-sea environments.

    There is a constant need to find feed additives that improve health and nutrition of farmed fish and lessen the intestinal inflammation induced by plant-based ingredients. The objective of this study was to evaluate the effects of adding an organic acid salt to alleviate some of the detrimental effects of extreme plant-ingredient substitution of fish meal (FM) and fish oil (FO) in gilthead sea bream diet. Three experiments were conducted. In a first trial (T1), the best dose (0.4%) of sodium butyrate (BP-70 ®NOREL) was chosen after a short (9-weeks) feeding period. In a second longer trial (T2) (8 months), four diets were used: a control diet containing 25% FM (T2-D1) and three experimental diets containing 5% FM (T2-D2, T2-D3, T2-D4). FO was the only added oil in D1, while a blend of plant oils replaced 58% and 84% of FO in T2-D2, and T2-D3 and T2-D4, respectively. The latter was supplemented with 0.4% BP-70. In a third trial (T3), two groups of fish were fed for 12 and 38 months with D1, D3 and D4 diets of T2. The effects of dietary changes were studied using histochemical, immunohistochemical, molecular and electrophysiological tools. The extreme diet (T2-D3) modified significantly the transcriptomic profile, especially at the anterior intestine, up-regulating the expression of inflammatory markers, in coincidence with a higher presence of granulocytes and lymphocytes in the submucosa, and changing genes involved in antioxidant defences, epithelial permeability and mucus production. Trans-epithelial electrical resistance (Rt) was also decreased (T3-D3). Most of these modifications were returned to control values with the addition of BP-70. None of the experimental diets modified the staining pattern of PCNA, FABP2 or ALPI.     The aquatic metabolic unit used in this study was cofounded by The Netherlands Organization for Scientific Research (code 805-34.025). In aquaculture, fish may be exposed to sub-optimal rearing conditions, which generate a stress response if full adaptation is not displayed. However, our current knowledge of several coexisting factors that may give rise to a stress response is limited, in particular when both chronic and acute stressors are involved. This study investigated changes in metabolic parameters, oxidative stress and innate immune markers in a rainbow trout (Oncorhynchus mykiss) isogenic line exposed to a combination of dietary (electrolyteimbalanced diet, DEB 700 mEq Kg−1 ) and environmental (hypoxia, 4.5 mg O2 L −1 ) challenges and their respective controls (electrolyte-balanced diet, DEB 200 mEq Kg−1 and normoxia, 7.9 or mg O2 L −1 ) for 49 days. At the end of this period, fish were sampled or subjected to an acute stressor (2 min of handling/confinement) and then sampled. Feeding trout an electrolyte-imbalanced diet produced a reduction in blood pH, as well as increases in cortisol levels, hepato-somatic index (HSI) and total energy content in the liver. The ratio between the lactate dehydrogenase (LDH) and isocitrate dehydrogenase (IDH) activities decreased in the liver of trout fed the DEB 700 diet, but increased in the heart, suggesting a different modulation of metabolic capacity by the dietary challenge. Several markers of oxidative stress in the liver of trout, mainly related to the glutathione antioxidant system, were altered when fed the electrolyteimbalanced diet. The dietary challenge was also associated with a decrease in the alternative complement pathway activity (ACH50) in plasma, suggesting an impaired innate immune status in that group. Trout subjected to the acute stressor displayed reduced blood pH values, higher plasma cortisol levels as well as increased levels of metabolic markers associated with oxidative stress in the liver. An interaction between diet and acute stressor was detected for oxidative stress markers in the liver of trout, showing that the chronic electrolyte-imbalance impairs the response of rainbow trout to handling/confinement. However, trout reared under chronic hypoxia only displayed changes in parameters related to energy use in both liver and heart. Taken together, these results suggest that trout displays an adaptative response to chronic hypoxia. Conversely, the dietary challenge profoundly affected fish homeostasis, resulting in an impaired physiological response leading to stress, which then placed constraints on a subsequent acute challenge. info:eu-repo/semantics/publishedVersion

    Infectious diseases and fish feeds management are probably the major expenses in the aquaculture business. Hence, it is a priority to define sustainable strategies which simultaneously avoid therapeutic procedures and reinforce fish immunity. Currently, one preferred approach is the use of immunostimulants which can be supplemented to the fish diets. Arginine is a versatile amino acid with important mechanisms closely related to the immune response. Aiming at finding out how arginine affects the innate immune status or improve disease resistance of European seabass (Dicentrarchus labrax) against vibriosis, fish were fed two arginine-supplemented diets (1% and 2% arginine supplementation). A third diet meeting arginine requirement level for seabass served as control diet. Following 15 or 29 days of feeding, fish were sampled for blood, spleen and gut to assess cell-mediated immune parameters and immune-related gene expression. At the same time, fish from each dietary group were challenged against Vibrio anguillarum and survival was monitored. Cell-mediated immune parameters such as the extracellular superoxide and nitric oxide decreased in fish fed arginine-supplemented diets. Interleukins and immune-cell marker transcripts were down-regulated by the highest supplementation level. Disease resistance data were in accordance with a generally depressed immune status, with increased susceptibility to vibriosis in fish fed arginine supplemented diets. Altogether, these results suggest a general inhibitory effect of arginine on the immune defences and disease resistance of European seabass. Still, further research will certainly clarify arginine immunomodulation pathways thereby allowing the validation of its potential as a prophylactic strategy.     The present study aimed to evaluate whether the total or high substitution of fsh meal (FM) and fsh oil (FO) by sustainable plant raw materials (plant meal and oils) in long-term feeding for rainbow trout, gilthead sea bream, and common carp can result in spoilage alterations during ice storage. Tese three species were fed throughout their whole rearing cycle with plant-based diets and compared to counterparts that received FM/FO-based diets or commercial-like diets. Sensory QIM schemes adopted for these species and ATP breakdown products (K-value and components) were used to evaluate the freshness. Sensory acceptability of 14, 15, and 12 days was found for rainbow trout, gilthead sea bream, and common carp, respectively. Tis corresponded to K-values of approximately 80%, 35%, and 65% for rainbow trout, gilthead sea bream, and common carp, respectively. No major efect of dietary history on postmortem shelf life was shown for gilthead sea bream and common carp; neither sensory-perceived nor chemical freshness showed diet-related diferences. Rainbow trout fed with the plant-based diet exhibited slightly worse sensory freshness than fsh fed with FM/FO-based diets, at the end of shelf life.     Background The Astrophorida (Porifera, Demospongiaep) is geographically and bathymetrically widely distributed. Systema Porifera currently includes five families in this order: Ancorinidae, Calthropellidae, Geodiidae, Pachastrellidae and Thrombidae. To date, molecular phylogenetic studies including Astrophorida species are scarce and offer limited sampling. Phylogenetic relationships within this order are therefore for the most part unknown and hypotheses based on morphology largely untested. Astrophorida taxa have very diverse spicule sets that make them a model of choice to investigate spicule evolution. Methodology/Principal Findings With a sampling of 153 specimens (9 families, 29 genera, 89 species) covering the deep- and shallow-waters worldwide, this work presents the first comprehensive molecular phylogeny of the Astrophorida, using a cytochrome c oxidase subunit I (COI) gene partial sequence and the 5′ end terminal part of the 28S rDNA gene (C1-D2 domains). The resulting tree suggested that i) the Astrophorida included some lithistid families and some Alectonidae species, ii) the sub-orders Euastrophorida and Streptosclerophorida were both polyphyletic, iii) the Geodiidae, the Ancorinidae and the Pachastrellidae were not monophyletic, iv) the Calthropellidae was part of the Geodiidae clade (Calthropella at least), and finally that v) many genera were polyphyletic (Ecionemia, Erylus, Poecillastra, Penares, Rhabdastrella, Stelletta and Vulcanella). Conclusion The Astrophorida is a larger order than previously considered, comprising ca. 820 species. Based on these results, we propose new classifications for the Astrophorida using both the classical rank-based nomenclature (i.e., Linnaean classification) and the phylogenetic nomenclature following the PhyloCode, independent of taxonomic rank. A key to the Astrophorida families, sub-families and genera incertae sedis is also included. Incongruences between our molecular tree and the current classification can be explained by the banality of convergent evolution and secondary loss in spicule evolution. These processes have taken place many times, in all the major clades, for megascleres and microscleres. Peer reviewed

    Gilthead seabream (Sparus aurata) tend to increase fat deposition during summer farming conditions in the Mediterranean, which may negatively affect productive performance and consumers’ quality perception of the final product. Therefore, this study evaluated the impacts of protein to lipid ratios in low fishmeal/fish oil diets on growth performance, body composition, feed conversion and nutrient utilization of seabream on-grown during summer temperature conditions. The experimental diets contained low levels of fishmeal, fish oil, and crude protein (39%), differing in crude lipid content: 16% (MF diet) or 12% (LF diet). A growth trial was performed with seabream (initial weight: 100 ± 7 g) from August to October (water temperature: 23.1 ± 2.2 ºC). A digestibility trial was also performed (at 23 ºC). Key performance indicators, whole-body composition and activities of digestive enzymes were evaluated at the end of the experiment (64 days). Low dietary lipid levels negatively affected lipid, energy, and amino acid digestibility, and as a result, fish fed the LF diet presented higher nitrogen faecal losses. Still, the decrease in nutrient digestibility was not related to dietary effects on the digestive enzyme activities. The experimental diets did not compromise the activity of pancreatic, gastric, and intestinal digestive enzymes nor feed utilization, but a slight growth impairment was observed in fish fed the LF diet, probably due to the lower amino acid and lipid digestibility. However, a potential benefit of this dietary treatment towards reducing fat accumulation in seabream during summer was observed. Nevertheless, the environmental impact of the nitrogen losses during seabream on-growing should be considered when estimating the sustainability of the production. This study demonstrated that the optimisation of diet formulations should account for the environmental conditions, especially in Mediterranean aquaculture, so the economic and environmental impacts may be correctly evaluated towards a more sustainable fish production. info:eu-repo/semantics/publishedVersion

    Understanding the interactions among demographic parameters, mating system and population dynamics is key to predict the response of populations to global change. The Mediterranean red coral is a precious octocoral suffering from population decline due to overfishing and warming-driven mass mortality events. While the demographic consequences of these two pressures are well characterized, little is known regarding their impact on population dynamics and evolution of red coral populations. The main objective of this study was to fill this gap focusing more particularly on mating pattern and genetic drift. Combining sibship and progeny arrays analyses, a genetic characterization of the red coral mating system was conducted. In addition, a synchronic approach was developed comparing mating patterns in two populations with contrasting demographic patterns: a pristine-like population and a declining population. The results show that polyandry is likely to be the norm in red coral. The similar patterns of genetic diversity between adults and larvae combined with the lack of differential reproductive success among putative fathers did not support significant sweepstakes effects during larval production. While instantaneous biparental inbreeding was detected, no long-term inbreeding was observed even in the declining population. Mating patterns and effective population sizes in the two populations were not statistically different. Nevertheless, a trend towards a slightly higher inbreeding and a lower number of breeders was observed in the declining population. Accordingly, we hypothesized that an increase in male gamete dispersal may buffer the increase of genetic drift expected in the declining population. This feedback between demographic decline and reproductive pattern may potentially take part in the long-term persistence of red coral populations.     The biology, ecology, and dynamics of the deep-sea teleost black scabbardfish in the northeast Atlantic are reviewed. The black scabbardfish is a commercial bathypelagic species found in the NE Atlantic mostly from Iceland to the Canary Islands and Western Sahara, at depths from 800 to 1300 m. The spatial structure of its population is still uncertain, although the existence of one single stock that migrates around the NE Atlantic driven by feeding and reproduction is the most likely hypothesis consistent with available data. This review is based on data from commercial fisheries off the Faroe Islands, Hatton Bank, the west of the British Isles, and Portugal (mainland, Azores, and Madeira) and from Icelandic and Scottish scientific surveys collected between 1988 and 2012. Spawning of black scabbardfish occurs around Madeira and the Canary Archipelagos during the last quarter of the year. According to the migratory hypothesis, eggs, larvae, and possibly juveniles move north to areas from south of Icelandic and Faroe Islands to the west of the British Isles where they remain for some years to feed and grow. Then, they move south to the area off mainland Portugal, where they reach the size of first maturity but do not reproduce, and later move further south to the spawning grounds. Further studies are needed to understand which of the environmental conditions prevailing around Madeira and the Canaries, but not elsewhere, allow this species to mature and subsequently reproduce. This review suggests that a multidisciplinary approach is required to confirm the spatiotemporal migration and habitats used by black scabbardfish populations in the NE Atlantic at different life stages. Otolith contour shape and microchemistry, fatty acids, carbon and nitrogen stable isotopes, as well as steroid hormones are proposed as promising alternative tools for responding to this challenge. © 2013 EDP Sciences, IFREMER, IRD.
