    In the present study, in addition to farmyard manure (FYM), cowpea was applied as green manure and faba bean as an intercrop in an organic greenhouse tomato crop, aiming to increase the levels of soil N. Three experiments (E1, E2, E3) were carried out, in which legumes were either noninoculated or inoculated with rhizobia alone or together with plant growth, promoting rhizobacteria. Inoculation of legumes with rhizobia considerably increased N2 fixation in E1 but had no impact on N2 fixation in E2 and E3. In E1, the application of cowpea decreased yield because it imposed a stronger nematode infection as the cowpea plants acted as a good host for Meloidogyne. However, in E2 and E3 the nematode infection was successfully controlled and the legumes significantly increased the tomato yield when inoculated in E2, irrespective of legume inoculation in E3. The total N concentration in the tomato plant tissues was significantly increased by legume application in E2 and E3, but not in E1. These results show that legumes applied as green manure can successfully complement N supply via FYM in organic greenhouse tomato, while legume inoculation with rhizobia can increase the amounts of nitrogen provided to the crop via green manure.

    Xylella fastidiosa subsp. pauca strain ‘De Donno’ has been recently identified as the causal agent of a severe disease affecting olive trees in a wide area of the Apulia Region (Italy). While insights on the genetics and epidemiology of this virulent strain have been gained, the complex network of interactions with the main susceptible host remains to be explored. A fundamental tool for understanding such interactions is the development of bacterial mutants for functional analysis of genes involved in the host recognition, pathogenicity and insect transmission. Experimental studies have demonstrated natural competence of X. fastidiosa in the uptake of exogenous genetic material, a feature exploited for site-specific introduction or deletion of genes through homologous recombination. Nevertheless, numerous studies have shown that several factors may affect X. fastidiosa transformation efficiency, including growth rate, twitching motility, sequence similarity, and the presence of Restriction-Modification systems that cleave incoming DNA. On this basis, two different plasmids containing the chromosomal replication origin (oriC) of X. fastidiosa and E. coli were used to transform X. fastidiosa ‘De Donno’ in order to produce a GFP-expressing and a knockout strain for the rpfF gene, a crotonase producing a diffusible signal factor (DSF), involved in the quorum-sensing system. Repeated attempts to exploit natural competence, introducing the donor plasmids into X. fastidiosa ‘De Donno’ failed, highlighting the critical role of genetic diversity in recombination performances of this pathogen. Conversely, GFP and RpfF- mutants were successfully obtained by co-electroporation in the presence of an inhibitor of the Type I R-M system, that had been proved to impact the stable acquisition of foreign DNA by X. fastidiosa subsp. fastidiosa. Availability of mutants for one of the most virulent strains of X. fastidiosa opens for new explorations of host-microbe interactions, important to elucidate mechanisms underpinning the differential responses recorded upon infections of different olive cultivars and toward the implementation of strategies to mitigate the impact of the disease.

    In the Mediterranean, habitat-forming corals often characterize essential fish habitats. While their distribution is sufficiently known for the western basin, few data are available from the Central-Eastern Mediterranean Sea (CEM). This study fills this gap supplying the largest dataset ever built on the geographical and bathymetric distribution of the most relevant habitat-forming corals (Eunicella cavolini, Eunicella verrucosa, Eunicella singularis, Leptogorgia sarmentosa, Paramuricea clavata, Corallium rubrum and Savalia savaglia) of the CEM. Information collected from different sources such as literature, citizen science, and from the World Wide Web (WWW) was combined. Videos published on the WWW provided additional information on the presence of fishing lines and signs of damage, as well as on the distribution of purple and yellow-purple colonies of Paramuricea clavata. The study highlighted the impressive amount of information that the WWW can offer to scientists, termed here as Web Ecological Knowledge (WEK). The WEK is constantly fuelled by internauts, representing a free, refreshable, long-term exploitable reservoir of information. A quick and easy method to retrieve data from the WWW was illustrated. In addition, the distribution of corals was overlapped to marine protected areas and to the distribution of environmental conditions suitable for coralligenous habitats, fragile biogenic Mediterranean structures hosting complex assemblages in need of strict protection. The collected data allowed identifying priority areas with high species diversity and sites that are impacted by fishing activities. Supplied data can correctly address conservation and restoration policies in the CEM, adding an important contribution to ecosystem-based marine spatial planning.

    [Main conclusion] Both AE and SIE rarity on La Palma show a clear spatial pattern, with hotspots of endemic rarity found at high elevations and in rare climates, presumably owing to geographical and climatic constraints and possibly anthropogenic pressure (e.g., land use, introduced herbivores, fire). Areas of high rarity estimates coincide with the distribution and extent of PAs on La Palma, especially since the recent addition of the Natura 2000 sites. The hypervolume approach is a promising tool to estimate species range sizes, and can be applied on all scales where point/plot data are available. [Methods] We recorded all present endemic vascular plant species in 1,212 plots covering the entire island. We calculated endemic rarity (corrected range-rarity richness for endemics) using a rarity estimation approach based on kernel density estimations (hypervolume approach). We performed a sensitivity analysis based on multiple linear regressions and relative importance estimations of environmental drivers to estimate the performance of the hypervolume-based rarity estimation compared to standard methods (occurrence frequency, convex hulls, alpha hulls). [Aim] Rarity—an important measure for conservation biogeography—can vary over many orders of magnitude. However, it is unclear which regional-scale abiotic conditions drive processes affecting rarity of endemic species on islands. To support conservation efforts, we (1) assess the main abiotic drivers of endemic rarity, (2) determine how well existing protected areas (PAs) coincide with hotspots of endemic rarity and (3) introduce and evaluate a new hypervolume-based rarity estimator. [Results] Climate variables (mean annual temperature, climatic rarity, precipitation variability) best explained archipelago endemic (AE) and single-island endemic (SIE) rarity. Existing PAs covered the majority of AE and SIE rarity, especially national and natural parks as well as the Natura 2000 sites. In our study system, hypervolumes performed better than standard measures of range size.     For insect-borne plant pathogens, transmission biology is of major importance in outlining the disease epidemiology. The characteristics of acquisition, persistence and transmission of X. fastidiosa ST53 by the spittlebug Philaenus spumarius, the main vector in Apulia, are not yet described; similarly, transmission competence of the potential vector Cicadella viridis, the most common sharpshooter in Europe, is unknown. In this perspective, two sets of experiments were performed in 2017 and 2018 to study: i) the kinetics of bacterial multiplication and persistence in P. spumarius and C. viridis; ii) the influence of temperature, season and age of P. spumarius adults on simulated epidemic progression on olive plants under indoor and outdoor conditions. For the kinetics experiments, following the acquisition, insects were serially transferred in groups of five to olive or periwinkle test plants. In simulated epidemic progression experiments, after the acquisition, groups of insects were isolated in cages with 16 olive seedlings for different inoculation periods. Acquisition and transmission rates were assessed by testing individual insects after inoculation and by testing recipient plants 6 and 10 months post-inoculation. Furthermore, acquisition and transmission of X. fastidiosa ST53 by C. viridis were tested through an in vitro acquisition system. Overall, about 900 insects and 170 plants were tested in kinetics experiments, while about 800 spittlebugs and 1,500 plants were studied in simulated epidemic progression experiments. Preliminary results for P. spumarius indicate: a) a higher acquisition efficiency in September than July; b) a lower acquisition efficiency from periwinkle compared with olive as source plants, but higher transmission efficiency to periwinkle compared with olive as recipient plants. Cicadella viridis was able to acquire and transmit X. fastidiosa following acquisition on artificial diet or periwinkle, although with low efficiency.

    Benthic foraminifera are an important component of the marine biota, but protocols for investigating their viability and metabolism are still extremely limited. Classical studies on benthic foraminifera have been based on direct counting under light microscopy. Typically, these organisms are stained with Rose Bengal, which binds proteins and other macromolecules, but does not allow discrimination between viable and recently dead organisms. The fluorescent in situ hybridization technique (FISH) represents a new and useful approach to identify living cells possessing an active metabolism. Our work is the first test of the suitability of the FISH technique, based on fluorescent probes targeting the 18S rRNA, to detect live benthic foraminifera. The protocol was applied on <I>Ammonia</I> group and Miliolids, as well as on agglutinated polythalamous (i.e., <I>Leptohalysis scottii</I> and <I>Eggerella scabra</I>) and soft-shelled monothalamous (i.e., <I>Psammophaga</I> sp. and saccamminid morphotypes) taxa. The results from FISH analyses were compared with those obtained, on the same specimens assayed with FISH, from microscopic analysis of the cytoplasm colour, presence of pigments and pseudopodial activity. Our results indicate that FISH targets only metabolically active foraminifera, and allows discerning from low to high cellular activity, validating the hypothesis that the intensity of the fluorescent signal emitted by the probe is dependent upon the physiological status of cells. These findings support the usefulness of this molecular approach as a key tool for obtaining information on the physiology of living foraminifera, both in field and experimental settings.

    Abstract Mediterranean islands face significant environmental challenges due to their high population density, reliance on imports, and water scarcity, exacerbated by increasing risks from climate change. Nature-based solutions (NbS) could address these challenges sustainably and with multiple benefits, but their uptake in policy and planning is limited, and stakeholder perspectives are conspicuously lacking from current research. Here, we report the results of a collaborative, multi-stakeholder exercise to identify priority knowledge needs (KNs) that could enhance the uptake of NbS in Mediterranean islands. We used a well-established iterative prioritisation method based on a modified Delphi process. This was conducted by the authors, environmental policy and practice stakeholders from across the Mediterranean islands, representing business, government, NGOs and research. We developed a long list of potential KNs through individual submissions, and prioritised them through voting, discussion and scoring. Excepting workshop discussion, all individual contributions were anonymous. We present the 47 resulting KNs in rank order, classified by whether they can be addressed by knowledge synthesis and further research, or demand action in policy and practice. The top priority KNs are i) a more precise definition of NbS, ii) which NbS are adapted to dry Mediterranean conditions? iii) how to increase the adoption and use of NbS in urban plans?, iv) how can buildings and built-up areas be modified to accommodate green infrastructure and v) cost-benefit analysis of urban green spaces. In collaboration with these stakeholders, our findings will determine future research strategies on NbS implementation in the Mediterranean islands.

    In the olive groves, the main factor regulating spittlebugs population level is the abundances of vegetables ground cover and its species composition. Control of the juvenile’s vector population by soil tilling to reduce the emergence of adults represents one of the most efficient and environmentally sustainable approach toward the control of the spread of Xylella fastidiosa in the infected area, or to reduce the risk of its establishment in the buffer zones or free areas. In order to identify alternative strategies for soil management in olive groves, for two consecutive years (2017 – 2018) we compared five different soil management techniques: (i) natural and undisturbed ground vegetation; (ii) soil tilling performed twice (in early winter and in spring when the majority of the nymphs were at the IV instar); (iii) soil tilling performed only in winter; (iv) sowing Lolium spp; (v) sowing Hordeum vulgare. Two surveys per year were carried out. In each thesis, of approx. 3,000m2, 30 subplots of 0.25 m2 randomly distributed were surveyed by counting the number of spittle/plants and number of specimens (P. spumarius and N. campestris)/spittle. Soil tilling performed in winter and spring, effectively reduced almost to zero the presence of both spittlebug species, with an Abbott’s index equal to 99.6%. Conversely, when soil tilling was performed only in winter, it proved to be effective for reducing N. campestris population (Abbott’s index ranged from 50% to 60%) but not for P. spumarius. Sowing in winter Lolium spp. and Hordeum vulgare reduced P. spumarius juvenile populations, with an Abbott’s index of 60% and 40%, respectively. Whereas, results were inconsistent for N. campestris: in 2017 on both Poaceae treatment, the juvenile populations were lower than on the undisturbed natural ground cover, with an Abbott index equal to 58% (Lolium) and 86% (Hordeum vulgare), while the population resulting mostly abundant during survey carried out in 2018. These data further support the effectiveness of mechanical interventions for reducing incidence of juvenile of spittlebugs, and that using poaceae species to replace the ground vegetation may contribute to lowering P. spumarius populations.

    The evaluation of the effects of conservation agriculture during the transition from conventional tillage to no-tillage requires numerous indicators to be considered. For this purpose, we monitored changes in a multi-parameter dataset during a three-year experiment that combined three tillage intensities (conventional tillage—CT; minimum tillage—MT; and no tillage—NT) with three soil covering managements (tillage radish cover crop, winter wheat cover crop and bare soil). Using a multivariate analysis, we developed a Relative Sustainability Index (RSI) based on 11 physical (e.g., bulk density and penetration resistance), chemical (e.g., soil organic carbon and pH) and biological soil properties (e.g., earthworm density) to evaluate cropping systems sustainability. The RSI was most affected by tillage intensity showing higher RSI values (i.e., better performances) in reduced tillage systems. Specifically, the RSI under NT was 42% greater than that of CT and 13% greater than that of MT. Soil covering had little impact on the RSI. Among the tested parameters, the RSI was increased most by saturated hydraulic conductivity (+193%) and earthworm density (+339%) across CT and NT treatments. Our results suggest that conservation agriculture and, particularly, reduced tillage systems, have the potential to increase farm environmental and agronomic sustainability.

    Despite the increasing recognition of the quantitative importance of Archaea in all marine systems, the protocols for a rapid estimate of Archaeal diversity patterns in deep-sea sediments have been only poorly tested yet. Sediment samples from 11 deep-sea sites (from 79°N to 36°N, at depths comprised from 469 to 5,571 m) were used to compare the performance of two different primer sets (ARCH21f/ARCH958r and ARCH109f/ARCH 915r) and three restriction enzymes (AluI, Rsa I, and HaeIII) for the fingerprinting analysis of Archaeal diversity using terminal-restriction fragment length polymorphisms (T-RFLP). In silico and experimental analyses indicated that different combinations of primer sets and restriction enzymes provided different values of benthic Archaeal ribotype richness and different Archaeal assemblage compositions. The use of the ARCH109f/ARCH 915r primer set in combination with AluI provided the best results (a number of ribotypes up to four folds higher than other combinations), suggesting that this primer set should be used in future studies dealing with the analysis of the patterns of Archaeal diversity in deep-sea sediments. Multivariate multiple regression analysis revealed that, whatever the T-RFLP protocol utilized, latitude and temperature explained most of the variance in benthic Archaeal ribotype richness, while water depth had a negligible role. © 2009 Springer Science+Business Media, LLC.

    In the present study, in addition to farmyard manure (FYM), cowpea was applied as green manure and faba bean as an intercrop in an organic greenhouse tomato crop, aiming to increase the levels of soil N. Three experiments (E1, E2, E3) were carried out, in which legumes were either noninoculated or inoculated with rhizobia alone or together with plant growth, promoting rhizobacteria. Inoculation of legumes with rhizobia considerably increased N2 fixation in E1 but had no impact on N2 fixation in E2 and E3. In E1, the application of cowpea decreased yield because it imposed a stronger nematode infection as the cowpea plants acted as a good host for Meloidogyne. However, in E2 and E3 the nematode infection was successfully controlled and the legumes significantly increased the tomato yield when inoculated in E2, irrespective of legume inoculation in E3. The total N concentration in the tomato plant tissues was significantly increased by legume application in E2 and E3, but not in E1. These results show that legumes applied as green manure can successfully complement N supply via FYM in organic greenhouse tomato, while legume inoculation with rhizobia can increase the amounts of nitrogen provided to the crop via green manure.
