    White lupin is a high-protein crop requiring drought tolerance improvement. This study focused on a genetically-broad population of 138 lines to investigate the phenotypic variation and genotype × environment interaction (GEI) for grain yield and other traits across drought-prone and moisture-favourable managed environments, the trait genetic architecture and relevant genomic regions by a GWAS using 9828 mapped SNP markers, and the predictive ability of genomic selection (GS) models. Water treatments across two late cropping months implied max. available soil water content of 60–80% for favourable conditions and from wilting point to 15% for severe drought. Line yield responses across environments featured a genetic correlation of 0.84. Relatively better line yield under drought was associated with an increased harvest index. Two significant QTLs emerged for yield in each condition that differed across conditions. Line yield under stress displayed an inverse linear relationship with the onset of flowering, confirmed genomically by a common major QTL. An adjusted grain yield computed as deviation from phenology-predicted yield acted as an indicator of intrinsic drought tolerance. On the whole, the yield in both conditions and the adjusted yield were polygenic, heritable, and exploitable by GS with a high predictive ability (0.62–0.78). Our results can support selection for climatically different drought-prone regions.

    Lipids, components of the plasma and intracellular membranes as well as of droplets, provide different biological functions related to energy, carbon storage, and stress responses. Bacterial species display diverse membrane composition that changes in response to the different environmental conditions. During plant–pathogen interactions, lipids might have roles in several aspects such as recognition, signal transduction, and downstream responses. Among lipid entities, free fatty acids (FFAs) and their oxidized form, the oxylipins, represent an important class of signaling molecules in host–pathogen perception, especially related to virulence and defense. In bacteria, FFAs (e.g., diffusible signaling factors) and oxylipins have a crucial role in modulating motility, biofilm formation, and virulence. In this study, we explore by LC-TOF and LC-MS/MS the lipid composition of Xylella fastidiosa subsp. pauca strain De Donno in pure culture; some specific lipids (e.g., ornithine lipids and the oxylipin 7,10-diHOME), characteristic of other pathogenic bacteria, were revealed. Nicotiana tabacum was used for testing the ability of this pathogen in producing such lipids in the host. Different lipid compounds present a clear distribution pattern within the infected plant tissues compared to the uninfected ones.

    The skin mucus of gilthead sea bream was mapped by one-dimensional gel electrophoresis followed by liquid chromatography coupled to high resolution mass spectrometry using a quadrupole time-of-flight mass analyzer. More than 2,000 proteins were identified with a protein score filter of 30. The identified proteins were represented in 418 canonical pathways of the Ingenuity Pathway software. After filtering by canonical pathway overlapping, the retained proteins were clustered in three groups. The mitochondrial cluster contained 59 proteins related to oxidative phosphorylation and mitochondrial dysfunction. The second cluster contained 79 proteins related to antigen presentation and protein ubiquitination pathways. The third cluster contained 257 proteins where proteins related to protein synthesis, cellular assembly, and epithelial integrity were over-represented. The latter group also included acute phase response signaling. In parallel, two-dimensional gel electrophoresis methodology identified six proteins spots of different protein abundance when comparing unstressed fish with chronically stressed fish in an experimental model that mimicked daily farming activities. The major changes were associated with a higher abundance of cytokeratin 8 in the skin mucus proteome of stressed fish, which was confirmed by immunoblotting. Thus, the increased abundance of markers of skin epithelial turnover results in a promising indicator of chronic stress in fish.

    This paper analyzes various classes of processes associated with the tempered positive Linnik (TPL) distribution. We provide several subordinated representations of TPL L\'evy processes and in particular establish a stochastic self-similarity property with respect to negative binomial subordination. In finite activity regimes we show that the explicit compound Poisson representations gives rise to innovations following Mittag-Leffler type laws which are apparently new. We characterize two time-inhomogeneous TPL processes, namely the Ornstein-Uhlenbeck (OU) L\'evy-driven processes with stationary distribution and the additive process determined by a TPL law. We finally illustrate how the properties studied come together in a multivariate TPL L\'evy framework based on a novel negative binomial mixing methodology. Some potential applications are outlined in the contexts of statistical anti-fraud and financial modelling.

    The full exploitation of Composite Fiber Reinforced Polymers (CFRP) in so-called green aircrafts design is still limited by the lack of adequate quality assurance procedures for checking the adhesive bonding assembly, especially in load-critical primary structures. In this respect, contamination of the CFRP panel surface is of significant concern since it may severely affect the bonding and the mechanical properties of the joint. During the last years, the authors have developed and tested an electronic nose as a non-destructive tool for pre-bonding surface inspection for contaminants detection, identification and quantification. Several sensors and sampling architectures have been screened in view of the high Technology Readiness Level (TRL) scenarios requirements. Ad-hoc pattern recognition systems have also been devised to ensure a fast and reliable assessment of the contamination status, by combining real time classifiers and the implementation of a suitable rejection option. Results show that e-noses could be used as first line low cost Non Destructive Test (NDT) tool in aerospace CFRP assembly and maintenance scenarios.

    Big Earth Data-Cube infrastructures are becoming more and more popular to provide Analysis Ready Data, especially for managing satellite time series. These infrastructures build on the concept of multidimensional data model (data hypercube) and are complex systems engaging different disciplines and expertise. For this reason, their interoperability capacity has become a challenge in the Global Change and Earth System science domains. To address this challenge, there is a pressing need in the community to reach a widely agreed definition of Data-Cube infrastructures and their key features. In this respect, a discussion has started recently about the definition of the possible facets characterizing a Data-Cube in the Earth Observation domain. This manuscript contributes to such debate by introducing a view-based model of Earth Data-Cube systems to design its infrastructural architecture and content schemas, with the final goal of enabling and facilitating interoperability. It introduces six modeling views, each of them is described according to: its main concerns, principal stakeholders, and possible patterns to be used. The manuscript considers the Business Intelligence experience with Data Warehouse and multidimensional "cubes" along with the more recent and analogous development in the Earth Observation domain, and puts forward a set of interoperability recommendations based on the modeling views.

    AbstractMetagenomics has made accessible an enormous reserve of global biochemical diversity. To tap into this vast resource of novel enzymes, we have screened over one million clones from metagenome DNA libraries derived from sixteen different environments for carboxylesterase activity and identified 714 positive hits. We have validated the esterase activity of 80 selected genes, which belong to 17 different protein families including unknown and cyclase-like proteins. Three metagenomic enzymes exhibited lipase activity, and seven proteins showed polyester depolymerization activity against polylactic acid and polycaprolactone. Detailed biochemical characterization of four new enzymes revealed their substrate preference, whereas their catalytic residues were identified using site-directed mutagenesis. The crystal structure of the metal-ion dependent esterase MGS0169 from the amidohydrolase superfamily revealed a novel active site with a bound unknown ligand. Thus, activity-centered metagenomics has revealed diverse enzymes and novel families of microbial carboxylesterases, whose activity could not have been predicted using bioinformatics tools.

    This paper presents a new approach to statistically characterize and simulate the wave climate under storm conditions. The methodology includes the joint selection of the parameters that identify storm events (significant wave height threshold, minimum storm duration and minimum interarrival time between consecutive storms) by means of hypothesis testing on the distribution functions of the number of storm events and the elapsing time between storms, providing an improved characterization of the parameters that define storm events. The main wave variables and their temporal dependence are characterized by non-stationary mixture distribution functions and a vector autoregressive model. This allows to adequately reproduce the random temporal evolution of storm events, crucial for the study of damage progression in maritime structures without the use of predefined geometries. The long-term time series of storm events and calm periods is obtained using copula functions which analyze the joint dependence of storm duration and interarrival time for separate climate intervals. The model is applied to hindcast data at a location of the Mediterranean sea close to the Granada coast in Spain to show its ability to reproduce wave storm conditions accounting for the time variability of the storminess. An example of application, using a large number of simulations and a damage progression model in a maritime structure, is presented.     Abstract The aim of this work is to analyze the performance of the Overtopping Breakwater for Energy Conversion (OBREC), developed by the team of the University of Campania and installed at prototype scale in the port of Naples. It is a multifunctional coastal structure aimed to protect harbors and produce energy, based on the overtopping principle. This device has been preliminary analyzed by means of experimental and numerical investigations. This contribution provides for the first time an integrated assessment of the OBREC hydraulic and structural performance, by means of the measurements collected at the prototype installation and of numerical modelling i.e. methodologies not affected by the scale effects. The numerical model, developed in the openFOAM environment, is calibrated on the field data gathered during a storm event and is then applied to extend the information related to the OBREC response under the typical wave climate. The results obtained are proposed in terms of overtopping discharge rates and pressures acting along the OBREC profile, under several sea states. Based on these results some recommendations and indications regarding the optimal cross section design are given to maximize the energy production, without compromising the structural stability, and to promote the device exploitability.

    The study of planktonic organisms during abrupt climatic variations of the last glacial period (Dansgaard-Oeschger oscillations, D-O) may reveal important insights on climatic, oceanographic and biological interactions. Here we present planktic foraminifera and coccolithophore data collected at the Ocean Drilling Program Site 963 (Sicily Channel), with a mean sampling resolution of respectively 43.5 and 98.9. yr, over the interval between 70,000 and 20,000. yr ago. The paleoenvironmental reconstruction suggests that three different scenarios can be seen across each D-O cycle: 1. oligotrophic surface water and a deep thermocline for the early Interstadials; 2. a Deep Chlorophyll Maximum and coccolithophore winter/spring blooming in the late Interstadials; 3. reduced productivity together with the shallowing of the nutricline depth during Stadials and Heinrich events. The unique mode of productivity dynamics is corroborated by comparing our paleoecological results with those published from high-resolution cores in the Alboran Sea clearly indicating reduced trophic levels during Stadials and Heinrich events. Finally, we argue that the density contrast between the Atlantic water inflow and subsurface water may have affected productivity dynamics in such a large area. The strong vertical density gradient may have hampered the vertical convection of the water column, producing a negative effect on biological productivity, especially during Stadial phases. © 2013 Elsevier B.V.

    White lupin is a high-protein crop requiring drought tolerance improvement. This study focused on a genetically-broad population of 138 lines to investigate the phenotypic variation and genotype × environment interaction (GEI) for grain yield and other traits across drought-prone and moisture-favourable managed environments, the trait genetic architecture and relevant genomic regions by a GWAS using 9828 mapped SNP markers, and the predictive ability of genomic selection (GS) models. Water treatments across two late cropping months implied max. available soil water content of 60–80% for favourable conditions and from wilting point to 15% for severe drought. Line yield responses across environments featured a genetic correlation of 0.84. Relatively better line yield under drought was associated with an increased harvest index. Two significant QTLs emerged for yield in each condition that differed across conditions. Line yield under stress displayed an inverse linear relationship with the onset of flowering, confirmed genomically by a common major QTL. An adjusted grain yield computed as deviation from phenology-predicted yield acted as an indicator of intrinsic drought tolerance. On the whole, the yield in both conditions and the adjusted yield were polygenic, heritable, and exploitable by GS with a high predictive ability (0.62–0.78). Our results can support selection for climatically different drought-prone regions.

    Lipids, components of the plasma and intracellular membranes as well as of droplets, provide different biological functions related to energy, carbon storage, and stress responses. Bacterial species display diverse membrane composition that changes in response to the different environmental conditions. During plant–pathogen interactions, lipids might have roles in several aspects such as recognition, signal transduction, and downstream responses. Among lipid entities, free fatty acids (FFAs) and their oxidized form, the oxylipins, represent an important class of signaling molecules in host–pathogen perception, especially related to virulence and defense. In bacteria, FFAs (e.g., diffusible signaling factors) and oxylipins have a crucial role in modulating motility, biofilm formation, and virulence. In this study, we explore by LC-TOF and LC-MS/MS the lipid composition of Xylella fastidiosa subsp. pauca strain De Donno in pure culture; some specific lipids (e.g., ornithine lipids and the oxylipin 7,10-diHOME), characteristic of other pathogenic bacteria, were revealed. Nicotiana tabacum was used for testing the ability of this pathogen in producing such lipids in the host. Different lipid compounds present a clear distribution pattern within the infected plant tissues compared to the uninfected ones.
