D.; Chust, T.; Krasnoselskikh, V.; Kretzschmar, M.; Lorfèvre, E.; Plettemeier, D.; Souček, J.; Steller, M.; Štverák, Š.; Trávníček, P.; Vaivads, A.; Horbury, T.S.; O’Brien, H.; Angelini, V.; Evans, V.;Context.The recently released spacecraft potential measured by the RPW instrument on board Solar Orbiter has been used to estimate the solar wind electron density in the inner heliosphere.Aims.The measurement of the solar wind’s electron density, taken in June 2020, has been analysed to obtain a thorough characterization of the turbulence and intermittency properties of the fluctuations. Magnetic field data have been used to describe the presence of ion-scale waves.Methods.To study and quantify the properties of turbulence, we extracted selected intervals. We used empirical mode decomposition to obtain the generalized marginal Hilbert spectrum, equivalent to the structure functions analysis, which additionally reduced issues typical of non-stationary, short time series. The presence of waves was quantitatively determined by introducing a parameter describing the time-dependent, frequency-filtered wave power.Results.A well-defined inertial range with power-law scalng was found almost everywhere in the sample studied. However, the Kolmogorov scaling and the typical intermittency effects are only present in fraction of the samples. Other intervals have shallower spectra and more irregular intermittency, which are not described by models of turbulence. These are observed predominantly during intervals of enhanced ion frequency wave activity. Comparisons with compressible magnetic field intermittency (from the MAG instrument) and with an estimate of the solar wind velocity (using electric and magnetic field) are also provided to give general context and help determine the cause of these anomalous fluctuations.

Transport in construction is responsible for up to 30% of freight movement in cities, with a subsequent impact in terms of pollutant emissions. Different solutions have been provided to alleviate the negative impact of freight transport related to construction activity, although no guidance has been provided for replicability in cities. One solution whose potential benefits are being studied with the support of policy makers is Construction Consolidation Centres (CCC). This paper proposes a method based on the Analytic Hierarchy Process (AHP) and provides an index based on 45 indicators to evaluate the suitability of the implementation of a CCC in terms of the pillars of sustainability in combination with a technical pillar. Three real construction projects were assessed in Luxembourg City, Paris, and Valencia. Two critical attributes were identified: the estimated operational costs of the construction company and the potential demand level in the area. The results of the analysis allow for: (i) the extraction of knowledge related to the sustainability of the construction project, and (ii) logistics to be integrated into the planning and design stages of the construction activity. Furthermore, a general structure is also proposed for integrating other construction solutions where CCC is not suitable. © 2021 by the authors. Licensee MDPI, Basel, Switzerland.

Hendriksen; Sünje Johanna Pamp; Alessandra De Cesare;An inter-laboratory proficiency test was organized to assess the ability of participants to perform shotgun metagenomic sequencing of cold smoked salmon, experimentally spiked with a mock community composed of six bacteria, one parasite, one yeast, one DNA, and two RNA viruses. Each participant applied its in-house wet-lab workflow(s) to obtain the metagenomic dataset(s), which were then collected and analyzed using MG-RAST. A total of 27 datasets were analyzed. Sample pre-processing, DNA extraction protocol, library preparation kit, and sequencing platform, influenced the abundance of specific microorganisms of the mock community. Our results highlight that despite differences in wet-lab protocols, the reads corresponding to the mock community members spiked in the cold smoked salmon, were both detected and quantified in terms of relative abundance, in the metagenomic datasets, proving the suitability of shotgun metagenomic sequencing as a genomic tool to detect microorganisms belonging to different domains in the same food matrix. The implementation of standardized wet-lab protocols would highly facilitate the comparability of shotgun metagenomic sequencing dataset across laboratories and sectors. Moreover, there is a need for clearly defining a sequencing reads threshold, to consider pathogens as detected or undetected in a food sample. Published version

However, unlike real-time PCR or serological assays, HTS datasets generated for pathogen detection do not easily provide yes/no answers. Rather, results of the taxonomic read assignment need to be assessed by trained personnel to gain information thereof. Proficiency tests are important instruments of validation, harmonization, and standardization. Within the European Union funded project COMPARE [COllaborative Management Platform for detection and Analyses of (Re-) emerging and foodborne outbreaks in Europe], we conducted a proficiency test to scrutinize the ability to assess diagnostic metagenomics data. An artificial dataset resembling shotgun sequencing of RNA from a sample of contaminated trout was provided to 12 participants with the request to provide a table with per-read taxonomic assignments at species level and a report with a summary and assessment of their findings, considering different categories like pathogen, background, or contaminations. Analysis of the read assignment tables showed that the software used reliably classified the reads taxonomically overall. However, usage of incomplete reference databases or inappropriate data pre-processing caused difficulties. From the combination of the participants' reports with their read assignments, we conclude that, although most species were detected, a number of important taxa were not or not correctly categorized. This implies that knowledge of and awareness for potentially dangerous species and contaminations need to be improved, hence, capacity building for the interpretation of diagnostic metagenomics datasets is necessary. However, routine use of WGS-based AST is hindered by the need for bioinformatics skills and knowledge of antimicrobial resistance (AMR) determinants to operate the vast majority of tools developed to date. By leveraging on ResFinder and PointFinder, two freely accessible tools that can also assist users without bioinformatics skills, we aimed at increasing their speed and providing an easily interpretable antibiogram as output. Methods - The ResFinder code was re-written to process raw reads and use Kmer-based alignment. The existing ResFinder and PointFinder databases were revised and expanded. Additional databases were developed including a genotype-to-phenotype key associating each AMR determinant with a phenotype at the antimicrobial compound level, and species-specific panels for in silico antibiograms. ResFinder 4.0 was validated using Escherichia coli (n = 584), Salmonella spp. (n = 1081), Campylobacter jejuni (n = 239), Enterococcus faecium (n = 106), Enterococcus faecalis (n = 50) and Staphylococcus aureus (n = 163) exhibiting different AST profiles, and from different human and animal sources and geographical origins. Results - Genotype-phenotype concordance was ≥95% for 46/51 and 25/32 of the antimicrobial/species combinations evaluated for Gram-negative and Gram-positive bacteria, respectively. When genotype-phenotype concordance was <95%, discrepancies were mainly linked to criteria for interpretation of phenotypic tests and suboptimal sequence quality, and not to ResFinder 4.0 performance. Conclusions - WGS-based AST using ResFinder 4.0 provides in silico antibiograms as reliable as those obtained by phenotypic AST at least for the bacterial species/antimicrobial agents of major public health relevance considered. The development of new climate change policies has increased the motivation to reduce energy use in buildings, as reflected by a stringent regulatory landscape. The construction industry is expected to adopt new methods and strategies to address such requirements, focusing primarily on reducing energy demand, improving process efficiency and reducing carbon emissions. However, the realisation of these emerging requirements has been constrained by the highly fragmented nature of the industry, which is often portrayed as involving a culture of adversarial relationships and risk avoidance, which is exacerbated by a linear workflow. Recurring problems include low process efficiency, delays and construction waste. Building information modelling (BIM) provides a unique opportunity to enhance building energy efficiency (EE) and to open new pathways towards a more digitalised industry and society. BIM has the potential to reduce (a) waste and carbon emissions, (b) the endemic performance gap, (c) in-use energy and (d) the total lifecycle impact. BIM also targets to improve the whole supply chain related to the design, construction as well as the management and use of the facility. However, the construction workforce is required to upgrade their skills and competencies to satisfy new requirements for delivering BIM for EE. Currently, there is a real gap between the industry expectations for employees and current training and educational programmes. There is also a set of new requirements and expectations that the construction industry needs to identify and address in order to deliver more informed BIM for EE practices. This paper provides an in-depth analysis and gap identification pertaining to the skills and competencies involved in BIM training for EE. Consultations and interviews have been used as a method to collect requirements, and a portfolio of use cases have been created and analysed to better understand existing BIM practices and to determine current limitations and gaps in BIM training. The results show that BIM can contribute to the digitalisation of the construction industry in Europe with adapted BIM training and educational programmes to deliver more informed and adapted energy strategies.

On the one hand, some EDCs are used as important pharmaceutical drugs that one would not want to dismiss. On the other hand, these pharmaceutical drugs are having off-target effects and increasingly significant exposure to the general population with unwanted health implications. Flutamide, one of the top pharmaceutical products marketed all over the world for the treatment of prostate cancer, is also a pollutant. Its therapeutic action mainly depends on targeting the androgen receptors and inhibiting the androgen action that is essential for growth and survival of prostate tissue. Currently flutamide is of concern with respect to its categorization as an endocrine disruptor. In this work we have developed a physiologically based pharmacokinetic (PBPK) model of flutamide that could serve as a standard tool for its human risk assessment. First we built the model for rat (where many parameters have been measured). The rat PBPK model was extrapolated to human where the re-parameterization involved human-specific physiology, metabolic kinetics derived from in-vitro studies, and the partition coefficient same as the rat model. We have harmonized the model by integrating different sets of in-vitro, in-vivo and physiological data into a PBPK model. Then the model was used to simulate different exposure scenarios and the results were compared against the observed data. Both uncertainty and sensitivity analysis was done. Since this new whole-body PBPK model can predict flutamide concentrations not only in plasma but also in various organs, the model may have clinical applications in efficacy and safety assessment of flutamide. The model can also be used for reverse dosimetry in the context of interpreting the available biomonitoring data to estimate the degree to which the population is currently being exposed, and a tool for the pharmaceutical companies to validate the estimated Permitted Daily Exposure (PDE) for flutamide.

The European Union (EU) initiative on the Digital Transformation of Health and Care (Digicare) aims to provide the conditions necessary for building a secure, flexible, and decentralized digital health infrastructure. Creating a European Health Research and Innovation Cloud (HRIC) within this environment should enable data sharing and analysis for health research across the EU, in compliance with data protection legislation while preserving the full trust of the participants. Such a HRIC should learn from and build on existing data infrastructures, integrate best practices, and focus on the concrete needs of the community in terms of technologies, governance, management, regulation, and ethics requirements. Here, we describe the vision and expected benefits of digital data sharing in health research activities and present a roadmap that fosters the opportunities while answering the challenges of implementing a HRIC. For this, we put forward five specific recommendations and action points to ensure that a European HRIC: i) is built on established standards and guidelines, providing cloud technologies through an open and decentralized infrastructure; ii) is developed and certified to the highest standards of interoperability and data security that can be trusted by all stakeholders; iii) is supported by a robust ethical and legal framework that is compliant with the EU General Data Protection Regulation (GDPR); iv) establishes a proper environment for the training of new generations of data and medical scientists; and v) stimulates research and innovation in transnational collaborations through public and private initiatives and partnerships funded by the EU through Horizon 2020 and Horizon Europe.

Here we combine scenarios from integrated assessment models with a forward-looking life-cycle assessment to explore how alternative technology choices in power sector decarbonization pathways compare in terms of non-climate environmental impacts at the system level. While all decarbonization pathways yield major environmental co-benefits, we find that the scale of co-benefits as well as profiles of adverse side-effects depend strongly on technology choice. Mitigation scenarios focusing on wind and solar power are more effective in reducing human health impacts compared to those with low renewable energy, while inducing a more pronounced shift away from fossil and toward mineral resource depletion. Conversely, non-climate ecosystem damages are highly uncertain but tend to increase, chiefly due to land requirements for bioenergy.

Magnetic field data have been used to describe the presence of ion-scale waves.Methods.To study and quantify the properties of turbulence, we extracted selected intervals. We used empirical mode decomposition to obtain the generalized marginal Hilbert spectrum, equivalent to the structure functions analysis, which additionally reduced issues typical of non-stationary, short time series. The presence of waves was quantitatively determined by introducing a parameter describing the time-dependent, frequency-filtered wave power.Results.A well-defined inertial range with power-law scalng was found almost everywhere in the sample studied. However, the Kolmogorov scaling and the typical intermittency effects are only present in fraction of the samples. Other intervals have shallower spectra and more irregular intermittency, which are not described by models of turbulence. These are observed predominantly during intervals of enhanced ion frequency wave activity. Comparisons with compressible magnetic field intermittency (from the MAG instrument) and with an estimate of the solar wind velocity (using electric and magnetic field) are also provided to give general context and help determine the cause of these anomalous fluctuations.

Transport in construction is responsible for up to 30% of freight movement in cities, with a subsequent impact in terms of pollutant emissions. Different solutions have been provided to alleviate the negative impact of freight transport related to construction activity, although no guidance has been provided for replicability in cities. One solution whose potential benefits are being studied with the support of policy makers is Construction Consolidation Centres (CCC). This paper proposes a method based on the Analytic Hierarchy Process (AHP) and provides an index based on 45 indicators to evaluate the suitability of the implementation of a CCC in terms of the pillars of sustainability in combination with a technical pillar. Three real construction projects were assessed in Luxembourg City, Paris, and Valencia. Two critical attributes were identified: the estimated operational costs of the construction company and the potential demand level in the area. The results of the analysis allow for: (i) the extraction of knowledge related to the sustainability of the construction project, and (ii) logistics to be integrated into the planning and design stages of the construction activity. Hendriksen; Sünje Johanna Pamp; Alessandra De Cesare;An inter-laboratory proficiency test was organized to assess the ability of participants to perform shotgun metagenomic sequencing of cold smoked salmon, experimentally spiked with a mock community composed of six bacteria, one parasite, one yeast, one DNA, and two RNA viruses. Each participant applied its in-house wet-lab workflow(s) to obtain the metagenomic dataset(s), which were then collected and analyzed using MG-RAST. A total of 27 datasets were analyzed. Sample pre-processing, DNA extraction protocol, library preparation kit, and sequencing platform, influenced the abundance of specific microorganisms of the mock community. Our results highlight that despite differences in wet-lab protocols, the reads corresponding to the mock community members spiked in the cold smoked salmon, were both detected and quantified in terms of relative abundance, in the metagenomic datasets, proving the suitability of shotgun metagenomic sequencing as a genomic tool to detect microorganisms belonging to different domains in the same food matrix. The implementation of standardized wet-lab protocols would highly facilitate the comparability of shotgun metagenomic sequencing dataset across laboratories and sectors. However, unlike real-time PCR or serological assays, HTS datasets generated for pathogen detection do not easily provide yes/no answers. Rather, results of the taxonomic read assignment need to be assessed by trained personnel to gain information thereof. Proficiency tests are important instruments of validation, harmonization, and standardization. Within the European Union funded project COMPARE [COllaborative Management Platform for detection and Analyses of (Re-) emerging and foodborne outbreaks in Europe], we conducted a proficiency test to scrutinize the ability to assess diagnostic metagenomics data. An artificial dataset resembling shotgun sequencing of RNA from a sample of contaminated trout was provided to 12 participants with the request to provide a table with per-read taxonomic assignments at species level and a report with a summary and assessment of their findings, considering different categories like pathogen, background, or contaminations. Analysis of the read assignment tables showed that the software used reliably classified the reads taxonomically overall. However, usage of incomplete reference databases or inappropriate data pre-processing caused difficulties. From the combination of the participants' reports with their read assignments, we conclude that, although most species were detected, a number of important taxa were not or not correctly categorized. This implies that knowledge of and awareness for potentially dangerous species and contaminations need to be improved, hence, capacity building for the interpretation of diagnostic metagenomics datasets is necessary. However, routine use of WGS-based AST is hindered by the need for bioinformatics skills and knowledge of antimicrobial resistance (AMR) determinants to operate the vast majority of tools developed to date. By leveraging on ResFinder and PointFinder, two freely accessible tools that can also assist users without bioinformatics skills, we aimed at increasing their speed and providing an easily interpretable antibiogram as output. Methods - The ResFinder code was re-written to process raw reads and use Kmer-based alignment. The existing ResFinder and PointFinder databases were revised and expanded. Additional databases were developed including a genotype-to-phenotype key associating each AMR determinant with a phenotype at the antimicrobial compound level, and species-specific panels for in silico antibiograms. ResFinder 4.0 was validated using Escherichia coli (n = 584), Salmonella spp. (n = 1081), Campylobacter jejuni (n = 239), Enterococcus faecium (n = 106), Enterococcus faecalis (n = 50) and Staphylococcus aureus (n = 163) exhibiting different AST profiles, and from different human and animal sources and geographical origins. Results - Genotype-phenotype concordance was ≥95% for 46/51 and 25/32 of the antimicrobial/species combinations evaluated for Gram-negative and Gram-positive bacteria, respectively. When genotype-phenotype concordance was <95%, discrepancies were mainly linked to criteria for interpretation of phenotypic tests and suboptimal sequence quality, and not to ResFinder 4.0 performance. Conclusions - WGS-based AST using ResFinder 4.0 provides in silico antibiograms as reliable as those obtained by phenotypic AST at least for the bacterial species/antimicrobial agents of major public health relevance considered. The development of new climate change policies has increased the motivation to reduce energy use in buildings, as reflected by a stringent regulatory landscape. The construction industry is expected to adopt new methods and strategies to address such requirements, focusing primarily on reducing energy demand, improving process efficiency and reducing carbon emissions. However, the realisation of these emerging requirements has been constrained by the highly fragmented nature of the industry, which is often portrayed as involving a culture of adversarial relationships and risk avoidance, which is exacerbated by a linear workflow. Recurring problems include low process efficiency, delays and construction waste. Building information modelling (BIM) provides a unique opportunity to enhance building energy efficiency (EE) and to open new pathways towards a more digitalised industry and society. BIM has the potential to reduce (a) waste and carbon emissions, (b) the endemic performance gap, (c) in-use energy and (d) the total lifecycle impact. BIM also targets to improve the whole supply chain related to the design, construction as well as the management and use of the facility. However, the construction workforce is required to upgrade their skills and competencies to satisfy new requirements for delivering BIM for EE. Currently, there is a real gap between the industry expectations for employees and current training and educational programmes. There is also a set of new requirements and expectations that the construction industry needs to identify and address in order to deliver more informed BIM for EE practices. This paper provides an in-depth analysis and gap identification pertaining to the skills and competencies involved in BIM training for EE. Consultations and interviews have been used as a method to collect requirements, and a portfolio of use cases have been created and analysed to better understand existing BIM practices and to determine current limitations and gaps in BIM training. The results show that BIM can contribute to the digitalisation of the construction industry in Europe with adapted BIM training and educational programmes to deliver more informed and adapted energy strategies.

On the one hand, some EDCs are used as important pharmaceutical drugs that one would not want to dismiss. On the other hand, these pharmaceutical drugs are having off-target effects and increasingly significant exposure to the general population with unwanted health implications. Flutamide, one of the top pharmaceutical products marketed all over the world for the treatment of prostate cancer, is also a pollutant. Its therapeutic action mainly depends on targeting the androgen receptors and inhibiting the androgen action that is essential for growth and survival of prostate tissue. Currently flutamide is of concern with respect to its categorization as an endocrine disruptor. In this work we have developed a physiologically based pharmacokinetic (PBPK) model of flutamide that could serve as a standard tool for its human risk assessment. First we built the model for rat (where many parameters have been measured). The rat PBPK model was extrapolated to human where the re-parameterization involved human-specific physiology, metabolic kinetics derived from in-vitro studies, and the partition coefficient same as the rat model. We have harmonized the model by integrating different sets of in-vitro, in-vivo and physiological data into a PBPK model. Then the model was used to simulate different exposure scenarios and the results were compared against the observed data. Both uncertainty and sensitivity analysis was done. Since this new whole-body PBPK model can predict flutamide concentrations not only in plasma but also in various organs, the model may have clinical applications in efficacy and safety assessment of flutamide. The model can also be used for reverse dosimetry in the context of interpreting the available biomonitoring data to estimate the degree to which the population is currently being exposed, and a tool for the pharmaceutical companies to validate the estimated Permitted Daily Exposure (PDE) for flutamide.

The European Union (EU) initiative on the Digital Transformation of Health and Care (Digicare) aims to provide the conditions necessary for building a secure, flexible, and decentralized digital health infrastructure. Creating a European Health Research and Innovation Cloud (HRIC) within this environment should enable data sharing and analysis for health research across the EU, in compliance with data protection legislation while preserving the full trust of the participants. Such a HRIC should learn from and build on existing data infrastructures, integrate best practices, and focus on the concrete needs of the community in terms of technologies, governance, management, regulation, and ethics requirements. Here, we describe the vision and expected benefits of digital data sharing in health research activities and present a roadmap that fosters the opportunities while answering the challenges of implementing a HRIC. For this, we put forward five specific recommendations and action points to ensure that a European HRIC: i) is built on established standards and guidelines, providing cloud technologies through an open and decentralized infrastructure; ii) is developed and certified to the highest standards of interoperability and data security that can be trusted by all stakeholders; iii) is supported by a robust ethical and legal framework that is compliant with the EU General Data Protection Regulation (GDPR); iv) establishes a proper environment for the training of new generations of data and medical scientists; and v) stimulates research and innovation in transnational collaborations through public and private initiatives and partnerships funded by the EU through Horizon 2020 and Horizon Europe.

Beyond greenhouse gas emissions, the power sector is also responsible for numerous other environmental impacts. Here we combine scenarios from integrated assessment models with a forward-looking life-cycle assessment to explore how alternative technology choices in power sector decarbonization pathways compare in terms of non-climate environmental impacts at the system level. While all decarbonization pathways yield major environmental co-benefits, we find that the scale of co-benefits as well as profiles of adverse side-effects depend strongly on technology choice. Mitigation scenarios focusing on wind and solar power are more effective in reducing human health impacts compared to those with low renewable energy, while inducing a more pronounced shift away from fossil and toward mineral resource depletion. Conversely, non-climate ecosystem damages are highly uncertain but tend to increase, chiefly due to land requirements for bioenergy.
