Summary: Given concerns about the ambition and effectiveness of current climate policies, a case has been made for the combination of demand side policies such as carbon pricing with supply side bans on fossil fuel extraction. However, little is known about their interplay in the context of climate stabilization strategies. Here, we present a multi-model assessment quantifying the effectiveness of supply side policies and their interactions with demand-side ones. We explore a variety of fossil fuel bans with four integrated assessment models and find that international supply side policies reduce carbon emissions but not at sufficient levels to stabilize temperature increase to well below 2°C. When combined with demand side policies, supply side policies reduce the required carbon price, dampen reliance on CO2 removal technologies, and increase investment in renewable energy. The results indicate the opportunity to integrate fossil fuel bans alongside price-based policies when exploring pathways to reach ambitious mitigation targets.

Members of the Campylobacter lari group are causative agents of human gastroenteritis and are frequently found in shellfish, marine waters, shorebirds, and marine mammals. Within a One Health context, we used comparative genomics to characterize isolates from a diverse range of sources and geographical locations within Europe and Australia and assess possible transmission of food, animal, and environmental isolates to the human host. A total of 158 C. lari isolates from Australia, Denmark, France, and Germany, which included 82 isolates from human stool and blood, 12 from food, 14 from domestic animal, 19 from waterbirds, and 31 from the environment were analyzed. Genome-wide analysis of the genetic diversity, virulence, and antimicrobial resistance (AMR) traits was carried-out. Most of the isolates belonged to C. lari subsp. lari (Cll; 98, 62.0%), while C. lari subsp. concheus and C. lari urease-positive thermotolerant Campylobacter (UPTC) were represented by 12 (7.6%) and 15 (9.5%) isolates, respectively. Furthermore, 33 (20.9%) isolates were not assigned a subspecies and were thus attributed to distant Campylobacter spp. clades. Whole-genome sequence-derived multilocus sequence typing (MLST) and core-genome MLST (cgMLST) analyses revealed a high genetic diversity with 97 sequence types (STs), including 60 novel STs and 14 cgMLST clusters (≤10 allele differences), respectively. The most prevalent STs were ST-21, ST-70, ST-24, and ST-58 (accounting for 13.3%, 4.4%, 3.8%, and 3.2% of isolates, respectively). A high prevalence of the 125 examined virulence-related loci (from 76.8 to 98.4% per isolate) was observed, especially in Cll isolates, suggesting a probable human pathogenicity of these strains. IMPORTANCE Currently, relatedness between bacterial isolates impacting human health is easily monitored by molecular typing methods. These approaches rely on discrete loci or whole-genome sequence (WGS) analyses. Campylobacter lari is an emergent human pathogen isolated from diverse ecological niches, including fecal material from humans and animals, aquatic environments, and seafood. The presence of C. lari in such diverse sources underlines the importance of adopting an integrated One Health approach in studying C. lari population structure for conducting epidemiological risk assessment. This retrospective study presents a comparative genomics analysis of C. lari isolates retrieved from two different continents (Europe and Australia) and from different sources (human, domestic animals, waterbirds, food, and environment). It was designed to improve knowledge regarding C. lari ecology and pathogenicity, important for developing effective surveillance and disease prevention strategies.

Smart monitoring has been studied and developed in recent years to create faster, cheaper, and more user-friendly on-site methods. The present study describes an innovative technology for investigative monitoring of heavy metal pollution (Cu and Pb) in surface water. It is composed of an autonomous surface vehicle capable of semiautonomous driving and equipped with a microfluidic device for detection of heavy metals. Detection is based on the method of square wave anodic stripping voltammetry using carbon-based screen-printed electrodes (SPEs). The focus of this work was to validate the ability of the integrated system to perform on-site detection of heavy metal pollution plumes in river catchments. This scenario was simulated in laboratory experiments. The main performance characteristics of the system, which was evaluated based on ISO 15839 were measurement bias (Pb 75%, Cu 65%), reproducibility (in terms of relative standard deviation: Pb 11–18%, Cu 6–10%) and the limit of detection (4 µg/L for Pb and 7 µg/L for Cu). The lowest detectable change (LDC), which is an important performance characteristic for this application, was estimated to be 4–5 µg/L for Pb and 6–7 µg/L for Cu. The life span of an SPE averaged 39 measurements per day, which is considered sufficient for intended monitoring campaigns. This work demonstrated the suitability of the integrated system for on-site detection of Pb and Cu emissions from large and medium urban areas discharging into small water bodies. Philaenus spumarius is a vector of Xylella fastidiosa, one of the most dangerous plants pathogenic bacteria worldwide. There is currently no control measure against this pathogen. Thus, the development of vector control strategies, like generalist predators, such as spiders, could be essential to limit the spread of this vector-borne pathogen. In this study, a polymerase chain reaction (PCR)-based approach was developed to principally detect DNA of P. spumarius in the spider’s gut. Accordingly, 20 primer pairs, targeting the mitochondrial cytochrome oxidase I (COI) and cytochrome b (cytB) genes, were tested for specificity, sensitivity, and efficiency in detecting P. spumarius DNA. Overall, two primer sets, targeting COI gene (COI_Ph71F/COI_Ph941R) and the cytB gene (cytB_ Ph85F/cytB_Ph635R), showed the highest specificity and sensitivity, being able to amplify 870 pb and 550 bp fragments, respectively, with P. spumarius DNA concentrations 100-fold lower than that of the DNA of non-target species. Among these two primer sets, the cytB_Ph85F/cytB_Ph635R was able to detect P. spumarius in the spider Xysticus acerbus, reaching 50% detection success 82 h after feeding. The feasibility of this primer set to detect predation of P. spumarius by spiders was confirmed in the field, where 20% of the collected spiders presented positive amplifications. This work was supported by the EU H2020 Research Project XF-ACTORS “Xylella fastidiosa Active Containment Through a multidisciplinary-Oriented Research Strategy” (Grant Agreement 727987) and to the Foundation for Science and Technology (FCT, Portugal) for financial support by national funds FCT/MCTES to CIMO (UIDB/00690/2020). COVID-19 has caused a global pandemic with considerable impact. Studies have examined the influence of socioeconomic status and air pollution on COVID-19 risk but in low detail. This study seeks to further elucidate the nuances of socioeconomic status, as defined by the Index of Multiple Deprivation (IMD), air pollution, and their relationship. We examined the effect of IMD and air pollution on the likelihood of testing positive for SARS-CoV-2 among 66,732 UKB participants tested for SARS-CoV-2 from 16 March 2020 through 16 March 2021. Logistic regression was performed controlling for age, sex, ancestry and IMD or air pollution in the respective models. IMD and its sub-scores were significantly associated with increased risk of testing positive for SARS-CoV-2. All particulate matter less than 2.5 μm (PM2.5), nitrogen oxide (NOx), and nitrogen dioxide (NO2) levels were associated with increased likelihood of testing positive for SARS-CoV-2. Measures of green space and natural environment around participants’ homes were associated with reduced likelihood of SARS-CoV-2. Socioeconomic status and air pollution have independent effects on the risk of testing positive for SARS-CoV-2. Green space and natural environment space in the proximity of people’s homes may mediate the effect of air pollution on the risk of testing positive for SARS-CoV-2.

Amyloodiniosis is a disease resulting from infestation by the ectoparasitic dinoflagellate Amyloodinium ocellatum (AO) and is a threat for fish species such as European sea bass (ESB, Dicentrarchus labrax), which are farmed in lagoon and land-based rearing sites. During the summer, when temperatures are highest, mortality rates can reach 100%, with serious impacts for the aquaculture industry. As no effective licensed therapies currently exist, this study was undertaken to improve knowledge of the biology of AO and of the host-parasite relationship between the protozoan and ESB, in order to formulate better prophylactic/therapeutic treatments targeting AO. To achieve this, a multi-modal study was performed involving a broad range of analytical modalities, including conventional histology (HIS), immunohistochemistry (IHC) and confocal laser scanning microscopy (CLSM). Gills and the oro-pharyngeal cavity were the primary sites of amyloodiniosis, with hyperplasia and cell degeneration more evident in severe infestations (HIS). Plasmacells and macrophages were localised by IHC and correlated with the parasite burden in a time-course experimental challenge. CLSM allowed reconstruction of the 3D morphology of infecting trophonts and suggested a protein composition for its anchoring and feeding structures. These findings provide a potential starting point for the development of new prophylactic/therapeutic controls.

Abstract The outbreak of SARS-CoV-2 and subsequent spread of the disease COVID-19 became classified as a pandemic in March of 2020, leading to global safety measures introduced to limit the impact of the virus. This combination of safety measures has become commonly referred to as “lockdown”. The associated industry and lifestyle changes led to reductions in the anthropogenic emission of atmospheric pollutants such as black carbon (BC), which is transported from the mid-latitudes into the Arctic during the winter and spring. Measurements of BC and other anthropogenic pollutants are of increasing importance in the Arctic due to the rapid warming observed there in the past few decades. It is believed that BC has a significant role in this warming, and so understanding the Arctic's response to reduced BC emissions at lower latitudes will provide insight into how future changes might mitigate further warming. Reductions in BC have been reported worldwide, and so in this study, the impact of these reductions on BC concentrations at the High Arctic site Villum Research Station was investigated. The effect was examined from March 2020, around when global lockdowns began, to June 2020, when the Arctic haze period ended and BC levels were once again low. Firstly, the Danish Eulerian Hemispheric Model (DEHM) was used to assess this impact on BC concentrations by adjusting global anthropogenic pollution emission inventories to simulate those observed during the lockdown period and comparing the results to a similar model run with standard emission inventories. Secondly, equivalent BC data from an aethalometer at Villum Research Station were analysed, comparing the concentrations during the lockdown period to both aethalometer data from previous years and DEHM results from the lockdown period. It was found that when adjusted DEHM emission inventories were introduced from the 1st of March, the model predicted a reduction in BC concentrations beginning on the 10th of March and reached a 10% reduction by the 1st of April. This reduction fluctuated around 10% until the end of the Arctic haze period. Aethalometer data did not show any significant change from previous years, and no concentration reduction could be concluded from its comparison with DEHM results. This is likely because the predicted reduction of 10% is smaller than both the inter-annual and intra-annual variability of measured BC concentrations at Villum.

Our paper emphasizes the importance of the kitchen layout in facilitating consumers' food hygiene practices. A significant correlation was found between the sink placement (inside or outside the kitchen) and hygienic practices during food handling based on a survey performed on consumers from ten European countries, indicating that those who had the sink in the kitchen were more likely to perform proper hygiene practices than those who have not. The self-reported practices were supported by observed practices in 64 households from five European countries. The observational study combined with the examination of kitchen layouts revealed that the kitchen work triangle with its apexes represented by the kitchen sink, cooking stove and refrigerator, which is recommended for ergonomic reasons by architects and designers, did not necessarily support food hygiene practices in kitchens. Cross-contamination events were associated with the sink – countertop distances longer than 1 m. Based on this, a new kitchen triangle with its apexes represented by the kitchen sink, working place (usually countertop) and cooking stove, with the distance between the sink and the working place less than 1 m is proposed to be used as norm in kitchen designs for combining ergonomics with safety. This triangle is proposedly named the food safety triangle and is aimed to mitigate the risks of foodborne illnesses by creating an arrangement that facilitates hygiene practices. This study is the first to highlight the importance of implementing the concept of food safety in the kitchen design based on significant correlations between kitchen equipment placement and consumers’ food safety practices. Highlights • Sink placement in kitchens correlates with self-reported food handling practices. • Sink placement is also correlated with observed cross-contamination events. • Kitchen layouts based on the work triangle do not support food hygiene practices. • A new triangle named food safety triangle is suggested for kitchens' organisation. • Sink – countertop distances ≤1 m favour consumers' food hygiene practices. Pruning residues can have a high quality as feedstock for energy purposes and are largely available in Europe. However, it is still an untapped resource. Such scarce use is due to the need to optimize their supply chain in term of collection machines and the associate cost of collection. A modular chipper (prototype PC50) for pruning harvest was developed. Such prototype is adaptable to various harvesting logistics and may produce a higher quality woodchip compared with the one produced by shredders available in the market. In this work, we tested the performance and quality of the product delivered by the prototype PC50 in various conditions and plant species, after a modulation of the machine settings (counter-rotating toothed rollers [CRR] speed), loading systems ([LS], either big bag or container), and knife types ([KT], either discontinuous hoe shaped knives or continuous helicoidal knives). To take into account of the covariates in the experiment (Cropping season and plant species), LSmeans were computed to have an unbiased estimate of the treatments means. The modulation of LS and KT scarcely affected the performance of the machine. In particular, the choice of the KT affected the field efficiency when the LS was a Tilting box but not a Big Bag. Whereas the continuous knife resulted in a 97% higher material capacity compared to hoe shape knives, the last of which the amount of short sized (<16 mm) fractions compared to helicoidal knives. No role of the CCR was found on the machine performance, but increasing CRR speed reduced the chip apparent bulk density and the fraction chips with a size <8 mm.
